The insert has an internal HindIII site about 180 bp from the vector EcoRI site. The insert also includes DdeI sites (2), TaqI sites (2), Sau3A1 istes (2), and a RsaI site. Restriction digests of the clone give the following sizes (kb): PstI--3.0, 0.75; EcoRI--3.8; HindIII--3.2, 0.6. Also available from the ATCC are pBYr5 (ATCC 45074) and pBYr5bSP (ATCC 45076) which contain the 0.75 kb PstI fragment. The former is cloned in pUC9 and the latter is also in pSP64, but in the opposite orientation. pBYr5aSP was constructed by random priming. RNA transcribed from the SP6 promoter is the virion sense strand, and will hybridize to (-) strand RNA. |
