The insert contains internal EcoRI, BamHI, AccI and HincII sites. Restriction digests of the clone give the following sizes (kb): BamHI/PstI--3.1, 0.86, 0.32; BamHI--3.1, 0.86, 0.32; PstI--3.1, 1.2; HindIII--4.2; SmaI--4.2. Linearize the plasmid with HindIII for in vitro transcription. This is a full-length clone of virus segment S11. The cDNA ends were tailored to BamHI and PstI sites before cloning into the PstI site of pGEM-1. |
