The 5' EcoRI site was lost in cloning. The insert contains the following sites (in approximate kb from the 5' end): AvaI--0.3; PvuII--0.5, XhoI--0.75; AvaI--0.8; PstI--1.15; BglI-1.35; XbaI--1.55; SacI--1.85. Restriction digests of the clone give the following sizes (kb): HindIII--6.5; EcoRI--6.5; PstI--4.6, 1.8; BamHI--6.5; SalI--6.5; HindIII/SstI--4.2, 1.2, 1.1. This insert codes for 480 residues of the collagenous region. It also encodes most of the COOH-terminal propeptide. This clone detects major mRNA species of about 5.1 and 4.6 kb in human skin fibroblasts and a minor transcript of about 4.4 kb. |
