Insert contains internal sites for HindIII (3), PstI (3), SalI (2), BamHI (2), AvaI (2) and EcoRI (1). Digestion of the plasmid with PstI yields fragments of 6.0, 2.5, 2.2, 1.1 kb. Restriction digests of the clone give the following sizes (kb): PstI--5.8, 2.3, 1.9, 1.2; BamHI--8.2, 3.1; EcoRI--6.2, 5.4; BamHI/EcoRI--4.0, 3.5, 2.8, 0.5; PvuII/AvaI--4.5, 2.8, 2.2, 1.7; HindIII--4.5, 2.6 doublet, 1.9. Mucoid colony morphology on agar supplemented with ampicillin (0.5 mg/ml). The entire streptokinase (skc) structural gene is on the 2.5 kb PstI fragment. Clone expresses streptokinase of Streptococcus equisimilis which is exported into the medium. |
