Restriction digests of the clone give the following sizes (kb): PstI--3.2, 2.3, 0.6, 0.55, 0.5, 0.3; SstI/SphI--4.2, 3.4; BamHI--4.6, 1.55, 1.4. pFRC11 includes the final nucleotide positions 3-3881. The 27 kDa ORF is encoded in sequences 123-831, the 57 kDa ORF from 926-2423, and the 37 kDa capsid ORF from 2427-3444. If there is a ribosomal frameshift, an 88 kDa fusion protein can be generated from 123-2423. The insert is oriented so that the transcript from the T7 promoter produces mRNA with positive polarity. |
