Restriction digests of the clone give the following sizes (kb): EcoRI--3.3, 1.7, 1.7, 1.7, 1.7, 1.7, 1.7; HindIII--3.3, 1.7, 1.7, 1.7, 1.7, 1.7, 1.7; EcoRI/HindIII--3.3, 1.7, 1.7, 1.7, 1.7, 1.7, 1.7; BamHI--14.4; PstI--14.4. EcoRI detects a 2.4 kb autosomal band and 5 Y-specific bands of 10 kb (A, maps to short arm), 7.5 kb (B, short arm), 6 kb (C, long arm), 4.5 kb (D, centromeric) and 1.7 kb (E, long arm). TaqI detects autosomal bands of 11 to 12 kb and 10.5 kb, and 4 Y-specific bands of 9 kb (equivalent to EcoRI band E), 8 kb (EcoRI band D), 3.5 kb (EcoRI band A or B) and 2.8 kb (two fragments, equivalent to EcoRI C and EcoRI A or B). Plasmid contains 6 copies of the 1.7 kb insert. Wash at high stringency (0.1X SSC, 68C). |
