The 5' PstI site was not regenerated during cloning. There are 2 PstI sites in the insert, so a PstI digest generates 124 bp and 1000 bp fragments from the insert. These represent all but 410 bp of the 5' region of the cDNA. Restriction digests of the clone give the following sizes (kb): EcoRI--5.9; HindIII--5.9; PstI--4.9, 1.1; BamHI--5.9. This cDNA includes the complete coding sequence plus 5' (12 nucleotides) and 3' (211 nucleotides) untranslated regions. |
