The insert includes the following restriction sites (in nucleotides from the 5' end of the published sequence): SmaI--580, PstI--638, SacI--1201. Restriction digests of the clone give the following sizes (kb): EcoRI/BamHI--2.9, 1.2; HindIII--3.9, 0.9; BamHI--4.1; PstI--3.15, 1.0; SalI--4.1; EcoRI/XbaI--2.9, 1.3; BamHI/XhoI--2.9, 1.3. The sequence was confirmed by partial sequence analysis and it includes and matches nucleotides 440 to 1649 of the published sequence. It encodes most of the protein coding region (437 - 1841). The insert is oriented such that linearizing with BamHI and using the T3 promoter gives the antisense strand, and linearizing with EcoRI and using the T7 promoter gives the sense strand. |
