Restriction digests analyzed on agarose gels give the following sizes (kb): Sst--5.1, 3.7, 2.8,0.4; XhoI--11.8; EcoRI--6.6, 5.4; EcoRI/Sst--4.4, 3.0, 2.0, 1.75, 0.56; EcoRI/SalI--6.0, 3.8, 1.8, 0.68. The 3' EcoRI/EcoRI fragment (5.4 kb) used in constructing this clone contains 3 of the 4 exons. Constructed from lambdac-fos(mouse)-2 (5' Sst/EcoRI) and pc-fos (mouse)-1 (3' EcoRI/EcoRI) to make a complete c-fos gene. The insert contains internal HindIII, XhoI, EcoRI, NcoI, SstI, SalI, and BamHI sites. |
