Restriction digests of the clone give the following sizes (kb): EcoRI--6.0, 3.55; BamHI--9.5; HindIII--9.5; AvaI--9.5; PvuII--6.8, 2.7. Contains 64 TaqI sites, 74 HinfI sites, 5 RsaI sites at 507, 822, 2610, 3095, 3176; 2 Sau3AI sites at 575, 2304; 1 HaeIII site at 3526, 1 ClaI site at 1067; and no MspI or SphI sites. Sequence is a tandem repeat of pentanucleotides; 229/713 were TTCCA and 298/713 were single nucleotide substitutions of that: 58-TTCGA, 33-TTCCT, 31-GTCCA, 27-TTCTA, 23-TTCAA, 22-TTGCA, 20-TTTCA. A 3.55 kb band is revealed in EcoRI, HaeIII, and StuI digests. TaqI and StuI digests seem to reveal polymorphisms but they are too dependent on hybridization conditions for definitive segregation studies. |
