Animals were immunized with purified CEC1-5 (soluble C-cadherin ectodomain) protein. Spleen cells were fused with Sp2/0 mouse myeloma cells. C-cadherin is the major cadherin expressed in the blastula and early gastrula and is largely responsible for holding cells in the embryo together. The AA5 antibody is the first antibody that can activate cadherin adhesive function. The AA5 antibody restores strong adhesion to activin-treated blastomeres. The C-cadherin-activating antibody strongly inhibits the elongation of animal caps, in response to activin, without affecting mesodermal gene expression. |

The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.

| Subculturing |
Protocol: Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 to 2 X 10(5) viable cells/ml. Interval: Maintain cell density between 5 X 10(4) and 8 X 10(5) viable cells/ml. Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density) |
