Restriction digests of the clone give the following sizes (kb): EcoRI--6.3; HindIII--6.3; PstI--4.4, 1.9. The insert includes 5' flanking sequences and exon 1. SstI polymorphism is about 10 kb 5' to exon 1 of POMC. Shows Mendelian segregation with codominance. Recorded as not polymorphic by Collaborative Research, meaning either not polymorphic in 0.8% agarose gels, or difficult to work with. Enzyme(s) not detecting polymorphism: BglII, PstI, BamHI, AvaII, BglI, MspI. Tested against 20 unrelated individuals. |
